<dc:description>Questa tesi di dottorato è incentrata sulla progettazione, la sintesi e la valutazione di nuovi agonisti selettivi per il recettore mu-opioide (MOR) che puntano a un sito di legame ortosterico alternativo. A differenza dei derivati convenzionali della morfina e dei composti a base di benzomorfano, che interagiscono esclusivamente con il sito ortosterico classico e sono associati a effetti collaterali come il reclutamento della beta-arrestina, il sito recentemente identificato offre il potenziale per un'attivazione terapeutica con effetti avversi ridotti (miosi, depressione respiratoria e tolleranza).&#xd;
Il progetto combina la modellazione molecolare come il docking e le simulazioni di dinamica molecolare per identificare un nuovo scaffold in grado di sostituire la struttura del benzomorfano. Sono stati incorporati dei sostituenti a base di amminoacidi per imitare le caratteristiche strutturali dei peptidi oppioidi endogeni. È stata sviluppata una procedura per la sintesi dei composti progettati e la loro sintesi è stata avviata. Questo fornisce una solida base per una futura valutazione farmacologica.</dc:description>
<dc:description>This PhD thesis focuses on the design, synthesis, and evaluation of novel selective agonists for the mu-opioid receptor (MOR) targeting an alternative orthosteric binding site. Unlike conventional morphine derivatives and benzomorphan-based compounds, which interact exclusively with the classical orthosteric site and are associated with side effects such as beta-arrestin recruitment, the newly identified site offers the potential for therapeutic activation with reduced adverse effects, including miosis, respiratory depression, and tolerance.&#xd;
The project combines molecular modeling such as docking and molecular dynamics simulations to identify a novel scaffold capable of replacing the benzomorphan structure. Amino acid-based substituents were incorporated to mimic the structural features of endogenous opioid peptides. A procedure for the synthesis of the designed compounds was developed and their synthesis was initiated. This provides a solid basis for future pharmacological evaluation.</dc:description>
