This work is part of the BRESOV project (https://bresov.eu), a breeding program in progress to enhance the biodiversity of cultivated and wild Brassica spp. for establishing new broccoli cultivars suitable for organic growing, with high qualitative traits to exploit in the frame of the new organic seed EU Directive 8484/2018. The parental lines were represented by 10 landraces of Brassica oleracea var. botrytis and B. oleracea var. italica, 8 wild relatives belonging to B. oleracea complex species (n=9), and 10 hybrid F1 produced between some landraces of violet cauliflower and wild species. The selected plants were self-pollinated and utilized for crossings and backcrossings, obtaining new HF1s that were bred in an organic field in 2018-19 and 2019-20. The selection was based on phenotypic features such as commercial harvesting period and flowering, size, colour, consistency, and compactness of the inflorescence. All the morphological characteristics of the HF1 genotypes were significantly different at p≤0.05, except for the main vegetative stem width, main vegetative stem length and petiole width. In particular, some HF1 hybrids showed a reduction in the number of days for inflorescence appearance, of about 6 days with respect to the parental lines. The breeding lines were characterized for sugars content (SSR by refractometry and total sugars by HPLC), ascorbic acid (AsA by HPLC), alkyl sulphides (by GC-MS), and antioxidant capacity (by DPPH assay). The accessions showed interesting variabilities (expressed as coefficient of variation, CV), with maximum CV for DPPH scavenging (79.7%) and minimum for SSR (5.8%). The sugars content ranged 11.6-36.5% dw (CV 27.4%). AsA content ranged 290-771 mg 100 g-1 dw, while DPPH index ranged 406-11308 mg eqAsA 100 g-1 dw. The two parameters were moderately correlated (r=0.582). Total alkyl-sulphides ranged 0.51-7.58 µmol g-1 dw, with a high CV (50.9%) and no correlation with the measured antioxidant index was found. The evaluated breeding lines showed very interesting quality traits for the organic production of Brassica spp. and of the related food chains.
