To determine the effects of lipids on experimental acute necrotizing pancreatitis (ANP) and any correlation between lipids and PGE2 prostaglandin synthesis, 120 rats were treated with lipids or indomethacin. ANP was induced in order to evaluate the modification of PGE2 tissue biosynthesis. During ANP PGE2 levels increased and this effect was enhanced by administration of lipids only in the groups where pancreatic phlogosis occurred. Lipid administration reduces the histological damage without a clear mechanism. Indomethacin treatment worsened the prognosis and the histological findings and significantly lowered the PGE2 tissue levels. These findings indicate that lipids could be administered during TPN in order to guarantee an adequate calorie intake without causing disagreeable side-effects whereas treatment with indomethacin should be carefully evaluated before clinical use.
